Abstract

ABSTRACT Commercial media are often costly and sometimes unavailable in resource-limited laboratories, making fungal identification difficult. The aim of this study is to formulate a selective culture medium using coconut, millet and soybean chaff for the growth of candida albicans. The Proximate analysis of the combined chaff carried out revealed carbohydrate content as (39.42%), crude fiber (20.45%), crude protein (16.85%), fat content (9.10%), ash (5.76%), moisture content (8.42%). The test organism (Candida albican) was identified and confirmed using Gram stain, biochemical and cultural characteristics. The medium was formulated using Fifty (50) gram of coconut, millet and soybean chaff combined in ratio of 20:15:15 respectively. It was boiled & extracted, and 100ml of the filtrate was used to formulate the culture medium, with 1g of agar agar added as the solidifying agent. The formulated medium and synthetic respectively (sabourand dextrose agar) was tested against Candida albicans. And the total viable count was recorded as 4.4x 10^6 cfu/mL & 4.8 x 10^6 cfu/mL for the formulated medium and synthetic (SDA). The results suggest that coconut, millet and soybean chaff can serve as a substitute for conventional media in fungal growth. Keywords: Candida albican, agro wastes, formulation , medium
