HISTOLOGICAL AND BIOCHEMICAL EFFECTS OF DIABETAMIN ON THE KIDNEY OF ALBINO RAT ALLOXAN INDUCED DIABETES
Abstract
ABSTRACT
The present study assessed the histological and biochemical effects of diabetamin on the kidney of albino rat alloxan induced diabetes. Twenty four (24) adult rats were used in this study. The animals were divided into four groups of six rats in each. They were all feed with rat pellet and clean water for the entire period of the project. The experimental animals which were Group 1,2and 3 was administered 130mg/kg of alloxan intraperitoneally to induce diabetes, diabetes was confirmed after blood samples taken after administration showed high glucose levels, the test animals were then treated with diabetmin for 14days by intubation. Group 1 had and an average weight of 0.113kg was administered 500mg, group 2 which had an average weight of 0.125kg was administered 1000mg while group 3 which had an average of 0.103kg was administered 250mg, treatment stopped for a period of 10days and the animals were sacrificed. The blood samples were taken for biochemical analysis while the kidney was fixed in 10% neutral buffered formalin, processed with the automatic tissue processing machine, sectioning was done at 3-5microns and section was stained with Haematoxylin and eosin (H & E) for light microscopy. Photomicrographs were then produced for group 1,2,3, and 4.However, There was a significant increase in the weight during this period(0.05),but when treatment stopped the blood glucose level increased again (p0.05) meaning it was not affected by the administration of the dosages. From the photomicrograph all the experimental animals had a normal histology when compared to the animal in the control group meaning that the induction and treatment with diabetmin had no effect on the histology of the kidney of the experimental animal. However, further investigation is however needed to establish how diabetmin reverse the effect of alloxan and whether or not it has an effect on the beta cells of the pancreas in short term and long term basis and if it can be used to treat type I diabetes mellitus.
