EFFECT OF MUCUNA PRURIENS FRACTIONS ON RELATIVE KIDNEYAND LIVER WEIGHT IN PHENYLHYDRAZINE INDUCED ANAEMIC RATS
Abstract
ABSTRACT
The present study is aimed at investigating the effect of Mucuna pruriens and its fractions on relative kidney and liver weight of phenylhydrazine induced anaemic rats. Thirty two (32) male wistar rats were assigned to eight (8) experimental groups (n=4). Group A-G, exception of Group H were induced with haemolytic anaemia following intraperitoneal injection of phenylhydrazine at 40 mg/kg for 2 days and treated orally for seven consecutive days as follows: Group A (A+CE 100mg/kg) and B (A+CE 200 mg/kg), received 100 and 200 mg/kg of crude extract of M.pruriens; Group C (A+MW 50 mg/kg)and D (A+MW 100 mg/kg) received 50 and 100 mg/kg of methanol:water; Group E (A+AM 50 mg/kg) and F (A+AM 100 mg/kg) received 50 and 100 mg/kg of Acetic acid:methanol; Group G (anaemic control) and Group H (non- anaemic control) received 0.38 ml/kg of normal saline. Rats of all groups were anaesthetized with intraperitoneal injection of ketamine (90mg/kg) and sacrificed 24 hours after the last treatment. The kidney and liver was homogenized in 1.5 of 0.9% sodium chloride (ice cold), the homogenate was centrifuged at 3500 rpm for 20 minutes and supernatant obtained, was used for the analysis of Relative kidney weight and liver weight. The results of Relative kidney weight significantly increased to 0.61±0.06 (%) and 3.82±0.20 (%) respectively in phenylhydrazine induced anaemic rats compared to normal control 0.56±0.05 (%) and 2.81±0.18 (%), However the observed increase in both Relative kidney weight and relative liver weight were ameliorated (p
