IMMUNOHISTOCHEMICAL EVALUATION OF MSH2 IN PROSTATE CANCER PATIENTS ATTENDING NIGER DELTA UNIVERSITY TEACHING HOSPITAL
Abstract
ABSTRACT
Prostate cancer is cancer of the prostate. The prostate is a gland in the male reproductive system that surrounds the urethra just below the bladder. DNA mismatch repair protein MSH2 also known as MutS homolog 2 or MSH2 is a protein that in humans is encoded by the MSH2 gene, which is located on chromosome 2. MSH2 is a tumor suppressor gene and more specifically a caretaker gene that codes for a DNA mismatch repair (MMR) protein. The aim of this study is to evaluate MSH2 in prostate cancer patients using immunohistochemical techniques. The study was carried out using 20 reported formalin fixed paraffin embedded archived prostate tissue blocks previously diagnosed of various pathologies as the study samples. Avidin Biotin complex (ABC) method also known as Avidin biotin immune-peroxidase method was applied on the formalin fixed paraffin embedded sections (FFPE). The result shows immunopositivity of MSH2 on prostate cancer cases studied. Out of 20 samples received sixteen 16 (80.0%) stained positive for MSH2 and 04 (20.0%) stained negative. In conclusion, we have shown that complex structural rearrangements in mismatch DNA repair genes MSH2 is experienced in prostrate of the patient studied.
