Abstract

This study aimed to investigate the effect of selenium supplementation on nitric oxide and angiotensin-converting enzyme levels in Wistar rats subjected to acetaminophen induced toxicity, thereby elucidating the potential protective mechanisms of selenium against drug-induced liver injury. Acetaminophen (ACT) is widely used as an analgesic and antipyretic drug, but excessive use can lead to drug-induced liver injury (DILI) through oxidative stress and inflammation. Selenium (SEL), a vital antioxidant, has been suggested to counteract oxidative damage. This study investigated the effects of selenium supplementation on nitric oxide (NO) and angiotensin-converting enzyme (ACE) levels in Wistar rats subjected to acetaminophen-induced toxicity, with a focus on the potential protective mechanisms of selenium against DILI. A total of twenty (20) Wistar rats of difference sex group where ramdomly divided into 4 groups, each containing 5 animals per group. Group one (control group) received normal rat chow and water only. Group two (Acetaminophen only) received same as group one and 0.2mls of 7 grams of acetaminophen same.Same as group 1 and 0.2mls of 7g of acetaminophen dissolved in 150mls of water. Group 3 (Selenium only) received same thing as group 1 and 0.2mls of 7g of selenium dissolved in 150mls of water. Group 4 (Selenium and Acetaminophen) received same as group 1, 2 and 3. After weighing the Administration of Selenium and Acetaminophen solution was done orally and each given the solution corresponding to its body weight. Wistar rats were divided into four groups: control, acetaminophen only (ACT), selenium only (SEL), and a co-administered selenium with acetaminophen group (SEL+ACT). Our study results revealed that acetaminophen significantly (P
