IMPACT OF CURCUMIN SUPPLEMENTATION ON PANCREATIC BETA CELL FUNCTION IN ALLOXAN-INDUCED DIABETIC MALE WISTAR RATS
Abstract
ABSTRACT
Alloxan, a toxic compound, is commonly used to induce experimental diabetes in animals by selectively damaging pancreatic beta cells, leading to impaired insulin secretion and hyperglycemia.Conversely, curcumin, a bioactive polyphenol extracted from turmeric, has been shown to exert protective and restorative effects in the human health. With a long history of traditional medicinal use, curcumin has been extensively studied for its therapeutic potential in managing various diseases, including diabetes, arthritis, and Alzheimer's disease. The Homeostatic Model Assessment for Beta-cell function (HOMA-B) is a widely used mathematical index that evaluates the functional capacity of pancreatic beta-cells, which produce insulin. This study investigated the impact of curcumin supplementation on pancreatic beta-cell function in alloxan-induced diabetic male Wistar rats.
Forty (45) male Wistar rats of average weight 160-200g were randomized into five (5) groups of nine (9) animals each in Group A, B, C, D and E. Group A (Control group) received water and feed ad libitum. Group B (DM group), rats weregiven diet and water for the first four weeks and alloxan (150mg/kg)was administered once to induce diabetics.Group C (DM + CUR) received alloxan induction afterward administered dissolved curcumin supplement (100mg/kg) (1ml) for four (4) weeks. Group D (DM +MET) this group received alloxan induction (150mg/kg) and then were confirmed with diabetics after 72 hours and were left for two (2) weeks followed by metformin (100mg/kg) administration in addition to diet and water Group E (CUR), this group received curcumin administration dissolved in olive oil for four weeks alongside diet and water. After the sixth week, the animals were anesthetized using chloroform and the pancreatic beta cell functions were evaluated. Statistical analysis of data was performed using Graph pad prism version 5.0 and comparison were made with One-way analysis of variance (ANOVA) following Tukey’s post hoc test to determine statistical significance. Results were expressed as mean ± SEM.
Results showed that alloxan induction caused a significantdecrease in the concentration of insulin and blood glucose level in DM (p
