THE IMPACT OF CURCUMIN SUPPLEMENTATION ON CARDIAC INJURY MARKERS IN DICHLORVOS EXPOSED MALE WISTAR RATS
Abstract
There has been an indiscriminate use of organophosphates pesticides (OP) such as dichlorvos (also known as DDVP) for agricultural and domestic purposes which has been reported to increase accidental poisoning and nonspecific multi-organ toxicity. Recently, dichlorvos has been reported to be a known risk factor for cardiotoxicity. Curcumin supplements have properties which could protect the heart against damages by toxicants. Thus, this study investigated the possible cardioprotective impacts of curcumin supplements on cardiac injury markers in dichlorvos exposed male Wistar rats.
Forty (40) male Wistar rats were randomly allocated into four (4) groups of ten (10) rats. Group A (control group) received only standard rat diet and water ad-libitum. Group B (DDVP group) rats were exposed to 2ml of DDVP via inhalation for 10 mins in a desiccator, daily for 2weeks. Group C (DDVP + Curcumin) rats were also exposed to 2ml of DDVP by inhalation in a desiccator for two weeks and then treated with 100 mg/kg of curcumin supplements dissolved in 1ml of olive oil for 4 weeks. Group D (Curcumin only) rats were pre-treated with curcumin only for 4 weeks.
After two (2) weeks of exposure and four (4) weeks of treatment, the rats were sacrificed after they have been anaesthetized using chloroform. The rats were then dissected and their heart were quickly dissected out and homogenised in phosphate buffer and centrifuged at 2000rpm for 5 mins at room temperature to obtain the supernatant for biochemical assay of lactate dehydrogenase (LDH), creatine kinase-myocardial band (CK-MB) and cardiac troponin I (cTnI). Statistical analysis of data was performed using Graph pad prism version 5.0 and comparison were made with One-way analysis of variance (ANOVA) following Tukey’s post hoc test to determine statistical significance. Result were expressed as mean ± SEM.
Exposure of rats to dichlorvos caused a significant increase (p
