EVALUATION OF AMELIORATIVE EFFECTS OF PYRIDOXINE ON TESTICULAR DAMAGE INDUCED BY EXTRACT OF MANIHOT ESCULENTA TUBER IN WISTAR RAT
Abstract
ABSTRACT
Manihot esculenta (cassava) is a staple food and key source of dietary energy in tropical regions, known for its resilience to drought and nutrient-poor soils, making it vital for agricultural production. The aim of the study was to evaluate ameliorative effects of pyridoxine on testicular damage induced by alcoholic extract of manihot esculenta tuber in wistar rat.
Twenty male Wistar rats, weighing between 100g, were randomly assigned into four groups (A-
D) with five rats in each group. Group A served as the control group and was fed rat feed and water ad libitum. Group B were administered 100mg/kg alcoholic extract of cassava tuber , while Group C were administered 200mg/kg alcoholic extract of cassava tuber . Group D received 200mg/kg alcoholic cassava extract alongside 50mg/kg pyridoxine as antioxidant. The experimental animals were weighed weekly and the result were analysed using one way analysis of variance(ANOVA). After the administration, the animals were weighed and sacrificed by cervical dislocation. Sperm samples were collected from caudal epididymis for semen analysis and the testicles were excised and fixed in Bouin's fluid for histological analysis.
The result on body weight analysis showed that all groups exhibited weight increase but not statistically significant. Semen analysis revealed that cassava extract alone reduced sperm count, motility, and morphology, whereas co-administration of pyridoxine in Group D improved sperm parameters, indicating a protective effect. Histological analysis showed normal testicular architecture in the control and Group B, while Group C exhibited poorly developed germinal cells and evidence of hemorrhagic changes. Group D displayed a mix of normal and impaired germinal cells, suggesting partial protective effects of pyridoxine.
Findings suggest that moderate doses of Manihot esculenta extract may not harm testicular structure while high doses may lead to damage, which can be mitigated by pyridoxine.
