Abstract

Methamphetamine is known to induce significant oxidative stress and inflammation in various tissues, including the lungs, leading to respiratory dysfunction. Vitamin D3 are recognized for their neuroprotective properties. This study investigates the impact of vitamin D3 on pulmonary oxidative stress markers in male Wistar rats exposed to methamphetamine . A total of 20 male wistar rats were divided into four groups (n=4 per group); control, methamphetamine group(10mg/kg),vitaminD3 group(1.5kg/mg) and methamphetamine(10mg/kg) plus vitamin D3(1.5/kg) treatment groups.. The study lasted for nine weeks. After administration, the rats were sacrificed by euthanasia process by injecting each rat with ketamine through the intraperitoneal route which caused them to sleep. Pulmonary tissue samples were excised, weighed and homogenized for biochemical analysis of oxidative stress markers, including malondialdehyde (MDA), glutathione (GSH), GST, GPX levels. Histopathological analysis was performed to evaluate structural changes in pulmonary tissues. Data were presented as mean ± standard deviation and one-way ANOVA analysis of variance followed by Tukey’s post-hoc test was used to assess significance , with the level of significance P < 0.05. Results indicated that methamphetamine exposure significantly decreased the body weight and increased relative pulmonary weight suggesting pulmonary hypertrophy, along with increased MDA levels and decreased GSH levels, highlighting enhanced oxidative stress in the lungs. Conversely, vitamin D3 treatment effectively mitigated these alterations, reducing MDA levels and restoring antioxidant enzyme activity. Histopathological examination revealed damage to pulmonary tissues in Meth, Vitamin D3 and Meth+Vitamin D3 treated groups when compared to the control group characterized by features of pneumonities v These findings suggest that vitamin D3 possess significant protective effects against pulmonary oxidative stress induced by methamphetamine exposure, decreasing oxidative stress markers and preserving pulmonary tissue structure. The findings underscore the ability of Vitamin D3 to combat respiratory complication induced by methamphetamine. Further investigation into its therapeutic potential in combating METH-related respiratory complications is needed.
