EVALUATION OF HISTOMORPHOLOGICAL AND BIOCHEMICAL CHANGES IN THE KIDNEY OF AGBO IBA TREATED ALBINO RATS
Abstract
This study investigated the effect of doses of Agbo iba herbal extract administered at different concentration on some biochemical parameters on the kidney and also the histology of the kidney. Thirty-Two (32) albino rats were used in this work and they were grouped into 4 groups A – Control, B, C and D (n=8).Group A (control): They were feed with growers’ marsh and water daily. Group B: They were feed with growers’ marsh and water daily and were administered 500mg/kg of Agbo iba herbal extract each orally by intubation. Group C: They were feed with growers’ marsh and water daily and were administered 700mg/kg of Agbo iba herbal extract each orally by intubation. Group D: They were feed with growers’ marsh and water daily and were administered 900mg/kg of Agbo iba herbal extract each orally by intubation. This research took place for 30 days, however on the 15th day half of the animals in each group were sacrificed and the organs harvested as well as the blood samples, which was sent to the laboratory for processing and analysis, the remaining set of animals were sacrificed on the 30th day. The animals were anaesthetized with chloroform and sacrificed, blood samples were collected and the organ (kidney) harvested, rinsed in normal saline and immediately fixed in 10% neutral buffered formalin, processed with the automatic tissue processing machine, sectioning was done at 3-5 microns and section was stained with Gill’s haematoxylin and eosin (H & E) for light microscopy and photomicrographs obtained. The evaluation of biochemical parameters on the kidney was not statistically significant (p>0.05) and all the results of the renal function test were within the control range, also noticed was the average weight of the rats that were sacrificed on the 30th day decreased, compared to the first set of animals that were sacrificed, this did not affect the experiment as the control group which was not administered any extract also reduced. The histological results from this research showed normal architecture renal capsules on the rats that were sacrificed on the 15th day, but compared to the other animals that were sacrificed on the 30th day and was administered high doses which resulted to injury of the glomeruli of the renal capsules and this could result to nephrotoxicity, therefore it is recommended that long term and high dose intake of Agbo iba herbal extract could result to renal injury and therefore be prevented.
