EFFECT OF AQUEOUS EXTRACT OF TAPINANTHUS GLOBIFERUS ON HISTOPATHOLOGY AND SOME KIDNEY INDICES IN SALT LOADED RATS
Abstract
This study evaluated the biochemical and histopathological effects of aqueous extract of Tapinanthus globiferus on kidney of the NaCl diet fed rat. Thirty (30) adult Spraque Dawley rats, weighing 150-200g, were used for the experiment. After one week acclimatization, the rats were divided into five groups of six rats each as follows; Group 1 (normal) received Normal rat chow and water daily, Group 2 (Negative control) received 8% salt diet and water daily, Group 3 (Positive control) received 8% salt diet + (40mg / kg captopril) daily, Group 4 received 8% salt diet+ (100 mg/kg) aqueous extract of Tapinanthus globiferus daily and Group 5 received 8% salt diet+ extract (200mg/kg) aqueous extract of Tapinanthus globiferus daily. On the last day, the rats were fasted overnight after which they were sacrificed and blood samples were collected from each rats into EDTA bottles, homogenate were prepared for further biochemical analysis. Antioxidant activities of prepared Tapinanthus globiferus was examined using Total phenol and Total flavonoid (in vitro) as well as Catalase activity and GSH quantification. The antihypertensive activities of the extracts were examined using, ACE and blood pressure measurement. Histopathology was conducted on the rat kidney tissue using hematoxylin and eosin staining method. From the result obtained, Total phenol and Total flavonoid contents of the aqueous extract of Tapinanthus globiferus increased with increase in concentration. There was a significant increase (p≤ 0.05) in total protein, catalase activity and GSH concentration. The activity of ACE was also reduced in the extract treated groups compared to the controls. The histopathology result of the kidney also showed improvements to the salt induced structural malformations in the extract treated rats compared to the negative control. Result from this study showed that the extract can be protective against salt induced organ damages and possible
hypertension.
