Abstract

ABSTRACT This study investigated the histological effects of ascorbate magnesium on the testicular architecture of ethanol-exposed Wistar rats. Chronic ethanol consumption is known to induce oxidative stress and reproductive toxicity, leading to structural and functional impairment of the testis. The present study aimed to evaluate the extent of testicular damage caused by ethanol and to determine the potential protective or restorative effects of ascorbate magnesium. A total of adult male Wistar rats were randomly divided into four groups: control, ethanol-treated, ascorbate magnesium-treated, and ethanol plus ascorbate magnesium-treated groups. Ethanol was administered orally to induce testicular toxicity, while ascorbate magnesium was administered to assess its protective effects. At the end of the experimental period, the animals were sacrificed, and the testes were harvested for histological examination using standard Hematoxylin and Eosin staining techniques. Results revealed that ethanol exposure caused significant reduction in body and testicular weights, along with marked histological alterations, including degeneration of seminiferous tubules, thinning of germinal epithelium, vacuolization, and reduced spermatogenic activity. In contrast, rats treated with ascorbate magnesium alone showed normal testicular architecture with active spermatogenesis. Notably, co-administration of ascorbate magnesium with ethanol resulted in significant improvement in testicular structure, evidenced by restoration of seminiferous tubules, increased germinal cell layers, and reduced cellular degeneration. These findings suggest that ethanol-induced testicular damage is largely mediated by oxidative stress, and that ascorbate magnesium exerts protective effects through its antioxidant properties. The study concludes that ascorbate magnesium has the potential to mitigate ethanol-induced reproductive toxicity and preserve testicular integrity.
