EFFECTS OF SYRINGIC ACID ON CISPLATIN-INDUCED NEPHROTOXICITY IN MALE WISTAR RATS.
Abstract
Syringic acid (SAC) is a known phenolic compound with anti-inflammatory and antioxidant actions. Due to these attributes, I investigated its prophylactic and therapeutic effects on cisplatin (CISP)-induced nephrotoxicity in male Wistar rats. The experimental design involved the control, CISP only, and SAC only groups administered normal saline, 5 mg/kg CISP intraperitoneally, and 50 mg/kg SAC orally. The prophylactic group received SAC (50 mg/kg) for 14 days before CISP (5 mg/kg) twice within 5 days. The therapeutic group received CISP first before SAC, as done in the prophylactic group. Compared with the control group, CISP-induced nephrotoxicity was characterized by a significant increase in the serum levels of potassium, sodium, BUN, uric acid, creatinine, kidney H2O2, TNF-α, NF-κB, Akt/PKB levels, and CAT activity. Also significantly decreased compared with the control were the kidney levels of PI3K, IL-6, Nrf2, GSH and activities of GPx, GST, and SOD. SAC treatments (both as a prophylactic and therapeutic agent) confer comparable nephroprotection against the effects of CISP. Although CISP toxicity was marked by oxidative stress in this study, our findings suggested that the mode through which SAC conferred nephroprotection against CISP-induced nephrotoxicity may be via anti-inflammatory mechanisms involving immunoenhancement through IL-6 upregulation and Akt-NF-κB-TNF-α downregulation in male Wistar rats.
