THE EFFECT OF HESPERIDIN ON THE CORTISOL LEVEL OF PSYCHOLOGICAL STRESS-INDUCED PTSD WISTAR RATS
Abstract
This study investigates the effect of hesperidin on the cortisol levels of psychological stress-induced PTSD Wistar rats following the administration of hesperidin to PTSD rats. This research aims to determine the effect of Hesperidin on the Cortisol level of psychological Stress-Induced PTSD in Adult Wistar Rats. Thirty healthy male Wistar rats weighing between 210-280g were divided into 6 groups: I, II, 11, IV, V, and VI with each group consisting of animals. Group I was regarded as the control group and took normal saline, Group 11 was regarded as the negative control group (stress-only group) and was not treated. Group III: stress + fluoxetine group, was administered 10mg/kg according to their body weight. Group IV: stress + hesperidin low dose, was administered hesperidin 50mg/kg. Group V: stress + hesperidin medium dose, was administered 100mg/kg. Group VI: received 200mg/kg respectively, and was treated once daily, all treatment was done orally using a 1ml syringe for 14 days. At the end of the administration period, all animals were Sacrificed using 20mg/kg of ketamine (intraperitoneal), and blood samples were collected for histological analysis were collected through the retro-orbital vein using a capillary tube. After a historical analysis was done and the Mean of Cortisol of the negative control (stress only 7.44+0.10) was compared against the mean of the treatment groups Group 3 (Stress + FXT, 6.75:0.04), Group 4 (Stress +Hesperidin LD, 6.67+0.06), Group 5 (Stress + Hesperidin MD 6.69+0.03) and Group 6 (Stress +Hesperidin HD 6.53+0.06) respectively, a significant difference (p0.05) was observed. The result shows a high mean value for cortisol in the stress-only group (7.44+0.10)and a lower mean value in the control group (6.19+0.13) and fluoxetine group (6.75+0.03). The hesperidin treatment groups show fluctuating mean values lesser than that of the stress group (Low 6.67+0.06; Medium 6.69+0.03; High 6.53:0.06). This result shows that hesperidin has a positive therapeutic effect on cortisol levels in PTSD induced Wistar rats.
